With recent increasing focus on the safety and activity of drug metabolites, it has become important to obtain authentic standards for biological testing and use as calibrants in bioanalytical methods.  However, many drug metabolites can be difficult or impossible to prepare using conventional synthetic organic chemistry.  In this short course methods for the biosynthesis and isolation of microgram quantities of drug metabolites will be described.  These un-weighable materials can be quantified using 1H-NMR spectroscopy to produce stock solutions of known concentration and purity.  Authentic standards prepared in this manner can then be used for in vitro pharmacological testing, measurement of enzyme kinetics, measurement of metabolite pharmacokinetics, and other uses.  Availability of these standards can open opportunities to carry out experiments that were previously unapproachable.    

The participant will learn:

a.       How to select a biological system for metabolite biosynthesis and carry out moderate-scale incubations;

b.      How to easily purify metabolites from these incubation mixtures;

c.       The application of various microbial systems to metabolite biosynthesis;

d.      Basics of NMR spectroscopy as applied to understanding metabolite structure;

e.       Use of NMR as a quantitative method to establish the concentration of stock solutions of biosynthesized metabolites at the microgram levels; and

f.       Applications of the uses of these materials in drug research. 

After attending this course, the participant should be able to return to their lab and generate metabolite standards to aid their research
